I heard him speak a few years ago at a dinner (No, not a Trekkie Convention!).  He said the sign came from a Jewish religious service he attended as a kid.  The Rabbi told everyone to bow their heads and close their eyes.  Young Leonard sneaked a peep at the Rabbi and saw that he had both hands raised making the now-famous sign.
