How do other providers pick which cities to tour?
As with anything in this biz, different ladies have different methods.  A good idea, IMO, is to start with cities that are familiar to you, or you want to visit them anyway, and hopefully where you've already had some requests/interest.

Do you do anything to test the waters by posting ads or do you just plan and hope there is enough interest?
Personally, I post ads and wait.  If I don't get enough pre-booked dates to cover basic travel expenses, I postpone or cancel.  This can be harder with some cities, like NYC & Chicago, that are well-known for having more last-minute booking guys. ;-)

Has anyone ever posted on the Regional Board to see ask for hobbiests to let them know how interested they would be? Is there anywhere to post a poll to ask on TER? If so, how do you do it?
Post on the new Travel board. Just a "What is there to do in Albuquerque in August?" kind of post should do it.  This will be perceived as a ThreAD, which it is, but from my understanding this is tolerated on that board. ;-)

One thing which I recommend: find travelling providers similar to yourselves and see what they're posting and where.  Use that as inspiration and a general how-to guide for your own posts/ads.  

Ads on message board sites are free, so start there first.  If you get a few nibbles, then pay for a Visiting Ad on Eros or other similar sites.

I cosign this 100%.  
Only thing I wanted to add was that I would recommend posting ads approximately 30 days out from the start of a your so you can gauge interest. I have seen/heard providers put up ads too early or too late. Make sure you utilize multiple channels and see what other providers recommend as marketing outlets for a particular city.
